Immunotherapy clinical studies should include extensive immune monitoring programmes to determine efficacy, investigate underlying immune mechanisms, and identify predictive biomarkers that may allow for patient stratification.

The immune monitoring unit of the VUMC-CCA Immunotherapy Lab has developed functional and multi-parameter phenotypic analyses for the immune profiling of patient-derived blood, tumour, and lymph node samples.

The combination of advanced flowcytometric analysis of on-treatment blood samples and of multiplex IHC analysis of pre- and post-treatment tumour biopsies will provide unique insights in the systemic and loco-regional dynamics of immune responses induced by the eTheRNA vaccine, as monotherapy, or combined with PD-1 blockade. Moreover, it may yield innovative and useful predictive biomarkers for future patient stratification.
